Measurement of Protein Dynamics from Site Directed Cu(II) Labeling

چکیده
The cover feature image illustrates two sites of a protein with different site specific reorientational dynamics (purple). Such differences can be measured by newly developed site-directed Cu(II) labeling methodology. resultant EPR lineshape at physiological temperatures is sensitive to the timescale backbone motion. Importantly, this methodology enables site-specific detection on both α-helices and β-sheets via EPR. Thus, role function elucidated. More information found in Review Sunil Saxena co-workers.

Site-directed nanoparticle labeling of cytochrome c.
Although nanoparticle-protein conjugates have been synthesized for numerous applications, bioconjugation remains a challenge, often resulting in denaturation or loss of protein function. This is partly because the protein-nanoparticle interface is poorly understood, which impedes the use of nanoparticles in nanomedicine. Although the effects of nanoparticle ligand and material on protein struct...
متن کاملSite-directed fluorescence labeling of a membrane protein with BADAN: probing protein topology and local environment.
The work presented here describes a new and simple method based on site-directed fluorescence labeling using the BADAN label that permits the examination of protein-lipid interactions in great detail. We applied this technique to a membrane-embedded, mainly alpha-helical reference protein, the M13 major coat protein. Using a high-throughput approach, 40 site-specific cysteine mutants were prepa...
متن کاملMapping membrane protein backbone dynamics: a comparison of site-directed spin labeling with NMR 15N-relaxation measurements.
The ability to detect nanosecond backbone dynamics with site-directed spin labeling (SDSL) in soluble proteins has been well established. However, for membrane proteins, the nitroxide appears to have more interactions with the protein surface, potentially hindering the sensitivity to backbone motions. To determine whether membrane protein backbone dynamics could be mapped with SDSL, a nitroxide...
متن کاملExamining the Conformational Dynamics of Membrane Proteins in situ with Site-directed Fluorescence Labeling
Two electrode voltage clamp electrophysiology (TEVC) is a powerful tool to investigate the mechanism of ion transport1 for a wide variety of membrane proteins including ion channels, ion pumps, and transporters. Recent developments have combined site-specific fluorophore labeling alongside TEVC to concurrently examine the conformational dynamics at specific residues and function of these protei...
